Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and intracellular transport. The Tau-430 isoform (2N4R) contains two N-terminal inserts and four microtubule-binding repeats, making it a key variant in adult human brain studies. The P290S mutation, located within the microtubule-binding domain, alters Tau's conformation and accelerates its aggregation into β-sheet-rich fibrils. Pre-formed fibrils (PFFs) generated from Tau-430 P290S mutant protein replicate early pathological features of tauopathies, including Alzheimer's disease and frontotemporal dementia. These fibrils mimic the structural and biochemical properties of neurofibrillary tangles observed in patient brains. Tau-430 P290S PFFs are used in cellular and animal models to investigate the mechanisms of Tau seeding, propagation, and neurotoxicity. Their consistent structure and disease relevance make them ideal for evaluating therapeutic strategies targeting Tau aggregation, clearance, and intercellular transmission. By modeling the transition from soluble Tau to pathogenic aggregates, Tau-430 P290S mutant PFFs provide a powerful platform for understanding tauopathy progression and accelerating the development of disease-modifying treatments.
