- Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and function. The Tau-441 isoform (2N4R), the longest variant expressed in the adult human brain, contains two N-terminal inserts and four microtubule-binding repeats, making it a key reference in tauopathy research. Under pathological conditions, wild-type Tau can misfold and aggregate into β-sheet-rich fibrils, contributing to the formation of neurofibrillary tangles observed in Alzheimer's disease and other tauopathies. Pre-formed fibrils (PFFs) generated from wild-type Tau-441 replicate early aggregation events and provide a physiologically relevant model for studying the mechanisms of Tau seeding, propagation, and neurotoxicity. Tau-441 wild-type PFFs are widely used in cellular and animal models to investigate the prion-like spread of Tau pathology, synaptic dysfunction, and neuronal degeneration. Their reproducibility and structural fidelity make them ideal for evaluating therapeutic strategies aimed at preventing aggregation, promoting clearance, and interrupting intercellular transmission.
